Jermall Charlo made his professional boxing debut on 12 August 2008, defeating Cimmaron Davis via technical knockout. He then defeated Deon Nash before meeting Carlos Garcia, who he also recorded victory against by a majority verdict.

Charlo won a boxing title, IBF light middleweight, for the first time on 12 September 2015 when he knocked out Cornelius Bundrage in the third round. He went on to defend the title three times against Wilky Campfort, Austin Trout and Julian Williams.

On 21 April 2018, Charlo faced Hugo Centeno Jr. for the vacant WBC interim middleweight title. He defeated Hugo via knockout. He has since defended the title against Matt Korobov, Brandon Adams, Dennis Hogan, Sergiy Derevyanchenko and Juan Macias Montiel.
