Nelson Semedo came through the youth system at Sintrense and made his professional debut at the age of 17. He joined Benfica on 12 January 2012, signing a five-year deal. He spent the 2012-13 season on loan at Fatima before returning to Benfica.

Semedo made his pro debut for Benfica on 10 August 2013 in a 0-0 draw against Trofense. He soon established himself as a regular starter in Benfica’s squad. The talented right-back signed a five-year deal afterwards and made his debut on 16 August. He scored his first competitive goal for the La Liga giants on 27 January 2019 in a 2-0 victory over Girona.
