Antonio Rudiger spent his youth career playing for local clubs in Germany. He began his football journey at VfB Sperber Neukolln in 2002 and later joined SV Tasmania Berlin. In 2005, he joined Neukollner Sportfreunde 1907 where he spent a season and the continued with his career at Hertha Zehlendorf.

Between 2008 and 2011, Rudiger played for Borussia Dortmund’s youth team. He then joined VfB Stuttgart and made his Bundesliga debut for the first team on 29 January 2012. He played 79 minutes as his side suffered a 3-0 defeat to Borussia Monchengladbach.
